A combination comprising a topoisomerase inhibitor and an anti-Trop-2 antibody, sacituzumab govitecan-hziy comprises the following three elements: the topoisomerase inhibitor SN-38; the humanized monoclonal antibody, hRS7 IgG1 (also known as sacituzumab), which binds to Trop-2; and • a hydrolysable linker (named CL2A), which connects the humanized monoclonal antibody to SN-38. Recombinant monoclonal antibodies are made by mammalian (murine myeloma) cells, whereas SN-38 and CL2A are made by chemical synthesis. Typically, sacituzumab govitecan-hziy comprises 7 to 8 SN-38 molecules per antibody molecule. Sacituzumab govitecan-hziy has a molecular weight of around 160 kilodaltons.
